My research group investigates molecular mechanisms underlying intracellular transport through the early secretory pathway. Our current focus is on the packaging of fully folded secretory proteins into coated vesicles that bud from the ER and the fusion of these vesicles with Golgi compartments. We study this process in the yeast Saccharomyces cerevisiae and in animal cell models.
Quantitative cell free assays that measure stages in ER-Golgi transport are used to dissect mechanisms through a combination of approaches including yeast molecular genetics, organelle isolation, protein purification and lipid biochemistry. We also monitor intracellular transport in vivo using live cell imaging and pulse/chase analyses.
