This is an outstanding review, chronologically advancing the understanding of growth-inhibitory factor (GIF) function and detailing its biochemical and physiological properties, including synaptic function and role as an oxidant scavenger.
The authors suggest in this mini-review that GIF, also referred to as metallothionine-3 (MT-3), while sharing metal-binding and reactive oxygen species scavenging properties with other MTs (MT-1 and MT-2), also shows distinct functions in mediating recovery upon neurodegeneration. The manuscript provides a state-of-the art account on the neuroprotective properties of GIF and the mechanisms related to its beneficial effects in Alzheimer’s disease (AD) and related pathologies. Yet, in a non-biased way, the authors also advance the hypothesis that GIF may also impede neuroregenerative processes. The mini-review highlights the need for a better understanding of the role of GIF in both healthy and disease conditions, a prerequisite for defining whether it may be potentially used as a therapeutic.

Neuronal growth-inhibitory factor, later renamed metallothionein-3, is one of four members of the mammalian metallothionein family. Metallothioneins are a family of ubiquitous, low-molecular-weight, cysteine-rich proteins. Although neuronal growth-inhibitory factor shares metal-binding and reactive oxygen species scavenging properties with the other metallothioneins, it displays several distinct biological properties. In this review, we examine the recent developments regarding the function of neuronal growth-inhibitory factor within the brain, particularly in response to brain injury or during neurodegenerative disease progression.
